  主要用途活化凝血因子5（FACTORVa）活性比色法定量檢測(cè)試劑是一種旨在通過活化凝血因子X（FXa）及其輔因子活化凝血因子（FVa），激活凝血酶，進(jìn)而水解多肽化合物底物H-D-Phe-Pip-Arg-pNA，釋放黃色對(duì)硝基苯胺，產(chǎn)生吸光峰值的變化，即采用比色法來測(cè)定血液樣品中活化凝血因子活性的權(quán)威而經(jīng)典的技術(shù)方法。該技術(shù)經(jīng)過精心研制、成功實(shí)驗(yàn)證明的。其適用于各種血漿樣品（動(dòng)物、人體等）活化凝血因子5的活性檢測(cè)。產(chǎn)品嚴(yán)格無菌，即到即用，操作簡(jiǎn)捷，性能穩(wěn)定。技術(shù)背景凝血因子5（FactorV）是維生素K非依賴性的不穩(wěn)定性單鏈糖蛋白，由肝組織、內(nèi)皮組織、巨核細(xì)胞、血小板等合成。血小板中含有20至25%凝血因子5。凝血因子5，作為活化凝血因子10的輔因子，成為凝血酶原復(fù)合物一員，包括活化凝血因子10、鈣離子和磷脂等，其功能在于轉(zhuǎn)化凝血酶原為凝血酶（THROMBIN；EC3.4.21.5），即活化凝血因子II（ActivatedFactorII；FactorIIa），構(gòu)成常見（common）凝血通路或機(jī)制。肝病、彌散性血管內(nèi)凝血（Disseminatedintravascularcoagulation；DIC）、原發(fā)性纖維蛋白溶解癥（primaryfibrinolysis）等引起凝血因子5減少或缺失。凝血因子5缺失將導(dǎo)致凝血酶原時(shí)間（prothrombintime；PT）和活化部分凝血活酶時(shí)間（activatedpartialthromboplastintime；APTT）延長(zhǎng)，而出現(xiàn)出血體質(zhì)。活化凝血因子X（FXa）及其輔因子活化凝血因子（FVa）一起，在鈣離子和磷脂的參與下，使凝血酶原（prothrombin）轉(zhuǎn)化為凝血酶（thrombin）基于由此水解人工合成的多肽化合物底物H-D-Phe-Pip-Arg-pNA（HD-苯丙氨酰-pipecolyl-精氨酰對(duì)硝基苯胺），釋放有色對(duì)硝基苯胺，在分光光度儀（405nm波長(zhǎng)）下，測(cè)定吸光峰值的變化，來定量測(cè)定活化凝血因子V的活性。其反應(yīng)系統(tǒng)為：[詳細(xì)]
