- Sensoryevaluationcanprovideintegrated,directmeasurementsoftheperceivedqualityoffoodproducts.However,asensorypanelissubjectiveandsuffersfrominconsistencyandinaccuracy.Inthispaper,weproposeasensoryevaluationsimulationmodelforLong極ngtea(aChinesebrandofgreentea).ThephysiochemicalqualityindicatorsofLong極ngteaweredeterminedbyinstrumentalanalysis,includingcolor,aroma,andtaste.Meanwhile,thesensoryqualityoftheteawasevaluatedbyanexpertsensorypanel.Anartificialneuralnetworkwasconductedtoapproximatelypredictsensoryevaluationscoresonthebasisofphysiochemicaldata.Theresultsshowedthatphysiochemicalfactors,includinghue,fluorescencepeak5,huechromascale,b,L,3-(methylthio)propionaldehyde,α-terpineol,linalool,dimethylsulfide,totalaromavalue,caffeine,quinicacid,theanin,gallicacidandtotalcatechinswerebestcorrelatedwithsensoryevaluationscores.Furthermore,physiochemicalfeaturesthatwerechosenaccordingtoimportantfactorweightswereusedtoclassifyLong極ngteaintotwogrades.Experimentalresultsdemonstratedthatinstrumentalanalysiscouldbecomplementarilyusedintheevaluationandcontrolofsensoryqualitybyestablishingareasonablesensory-instrumentcorrelationandhuman-simulatedpredictivemodel.[詳細]

- ThehumanpathogenicfungusCryptococcusneoformanshasalargepolysaccharide(PS)capsuleacopiousamountsofPSintoculturesandinfectedtissues.ThecapsularPSisamajorvirulencefactelicitprotectiveantibodyresponses.PSrecoveredfromculturesupernatantshashistoricallyprovidandconvenientsourceofmaterialforstructuralandimmunologicalstudies.TwomajorassumptionstudiesarethatthestructuralfeaturesoftheexopolysaccharidematerialfaithfullymirrorthoseofcthattheisolationmethodsdonotchangePSproperties.However,acomparisonofexopolysaccharitwoisolationtechniqueswithcapsularPSstrippedfromcellswithgammaradiationordimethylsurevealedsignificantdifferencesinglycosylcomposition,mass,size,charge,viscosity,circular-dichandreactivitywithmonoclonalantibodies.Ourresultsstronglysuggestthatexopolysaccharidesanarestructurallydifferent.AnoteworthyfindingwasthatPSmadebycetyltrimethylammoniumbroprecipitationhadalargermassandadifferentconformationthanPSisolatedbyconcentrationandsuggestingthatthemethodmostcommonlyusedtopurifyglucuronoxylomannanaltersthePS.HemethodusedtoisolatePScansignificantlyinfluencethestructuralandantigenicpropertiesofthepfindingshaveimportantimplicationsforcurrentviewsoftherelationshipbetweencapsularPSandexopolysaccharides,forthegenerationofPSpreparationssuitableforimmunologicalstudies,andfformulationofPS-basedvaccinesforthepreventionofcryptococcosis.[詳細]

- Electrostaticinteractionsbetweensyntheticpolyelectrolytesandproteinscanleadtotheformationofdense,macroion-richliquidphases,withequilibriummicroheterogeneitiesonlengthscalesuptohundredsofnanometers.TheeffectsofpHandionicstrengthontherheologicalandopticalpropertiesofthesecoacervatesindicatemicrostructuressensitivetoprotein-polyelectrolyteinteractions.Wereporthereonthepropertiesofcoacervatesobtainedforbovineserumalbumin(BSA)withthebiopolyelectrolytechitosanandfindremarkabledifferencesrelativetocoacervatesobtainedforBSAwithpoly(diallyldimethylammoniumchloride)(PDADMAC).CoacervationwithchitosanoccursmorereadilythanwithPDADMAC.Viscositiesofcoacervatesobtainedwithchitosanaremorethananorderofmagnitudelargerand,unlikethosewithPDADMAC,showtemperatureandshearratedependence.Forthecoacervateswithchitosan,afastrelaxationtimeindynamiclightscattering,attributabletorelativelyunrestrictedproteindiffusioninbothsystems,isdiminishedinintensitybyafactorof3-4,andtheconsequentdominancebyslowmodesisaccompaniedbyamoreheterogeneousarrayofslowapparentdiffusivities.Inplaceofasmall-angleneutronscatteringGuinierregioninthevicinityof0.004-1,a10-foldincreaseinscatteringintensityisobservedatlowerq.Takentogether,theseresultsconfirmthepresenceofdensedomainsonlengthscalesofhundredsofnanometerstomicrometers,whichincoacervatespreparedwithchitosanarelesssolidlike,moreinterconnected,andoccupyalargervolumefraction.Thedifferencesinpropertiesarethuscorrelatedwithdifferencesinmesophasestructure.[詳細]
