- 銀氨染色法： 1、取二塊雙向蛋白凝膠，用500mL固定液（40%甲醇+10%乙酸）固定30min 2、用雙蒸水沖洗3次，時(shí)間分別為3min、5min和3min 3、然后用500mL AgNO3染色液（500mL 中含有3.6% NaOH 10.5mL、20% AgNO3 9 mL和5mL氨水）染色32-33min 4、用雙蒸水沖洗4次，時(shí)間分別為3min、4min、4min和1min 5、然后用500mL顯色液（500mL中含有1%檸檬酸2.5mL和甲醛250μL）顯色5-12min（顯色時(shí)應(yīng)搖動(dòng)，顯色時(shí)間可靈活，一般采用邊顯色邊觀察） 6、500mL 5%醋酸終止反應(yīng)5min，然后用ddH2O沖洗2-3次 注意：雙向銀染上樣蛋白總量為40-250μg。

- 我的配方如下，僅供參考： 1. 固定：30min或過(guò)夜 100ml 乙醇 40% 乙醇 25ml冰醋酸 10% 冰醋酸 加水到250ml 2. 致敏：30min 300ml 乙醇 30% 乙醇 68g 乙酸鈉（或112.8g 三水乙酸鈉） 2g硫代硫酸鈉 加水到終體積1000ml 3. 水洗：3 x 5min 4. 銀染：20min 0.625g AgNO3 加水到終體積250ml 5. 水洗： 2 x 1 min (準(zhǔn)確掌握時(shí)間，水洗時(shí)間長(zhǎng)顯色速度慢，點(diǎn)的顏色淺) 6. 顯色：視情況而定 6.25 g Na2CO3 100 ul 37% 甲醛 （在使用前加入） 可加入 1g/L Na2S2O3 50微升以降低背景（在使用前加入） 加水到終體積250ml Sweet silver: 硼酸 3.1克 100 mM NaOH 3 克 150 mM 加水到終體積500 ml。 用前取sweet 225 ml加1g/L Na2S2O3 200微升，40%葡萄糖25 ml。 7. 終止：10min 7.3g EDTA.Na2.2H2O 或者2g 甘氨酸 加水到終體積500ml 8. 保存：1% 冰醋酸，4 ℃
