主要用途植物磷脂酶D(PhospholipaseD)總水解活性酶連續(xù)循環(huán)比色法定量檢測(cè)試劑是一種旨在通過(guò)磷脂酶D、脂酶、甘油-3-磷酸氧化酶和過(guò)氧化酶酶連續(xù)反應(yīng)系統(tǒng)中產(chǎn)生的紅色產(chǎn)物醌亞胺染料,在分光光度儀下峰值的,即采用比色法來(lái)測(cè)定植物組織裂解樣品中酶活性的權(quán)威而經(jīng)典的技術(shù)方法。該技術(shù)經(jīng)過(guò)精心研制、成功實(shí)驗(yàn)證明的。其適合于各種植物組織,包括種子(seed)、葉片(leaf)、根(root)、胚胎葉(cotyledon)、上胚軸(epicotyl)等裂解萃取樣品中磷脂酶D的總水解活性檢測(cè)。產(chǎn)品嚴(yán)格無(wú)菌,即到即用,操作簡(jiǎn)捷,性能穩(wěn)定。技術(shù)背景磷脂酶(phospholipase),是一種脂質(zhì)水解酶,催化磷脂的水解,產(chǎn)生脂肪酸和其它親脂產(chǎn)物,參與磷脂代謝。磷脂酶家屬有四種類(lèi)型亞酶,包括A、B、C和D。磷脂酶A(phospholipaseA;PLA)主要水解磷脂sn-1(磷脂酶A1催化)和sn-2(磷脂酶A2催化)位置上的乙酰基;磷脂酶B(phospholipaseB;PLB)同時(shí)水解磷脂的sn-1和sn-2位置上的乙?;?,又稱(chēng)為溶血磷脂酶(lysophospholipase);磷脂酶C(phospholipaseC;PLC)在磷酸基前水解,釋放二?;视停╠iacylglycerol;DAG)和第二信使分子三磷酸肌醇(inositoltriphosphate);磷脂酶D(phospholipaseD;PLD;EC3.1.4.4)在磷酸基后水解,釋放磷脂酸(phosphatidicacid)。植物磷脂酶D,屬于異源性酶(heterogeneousenzyme)家屬成員,與哺乳動(dòng)物磷脂酶D不同,有6個(gè)異構(gòu)體:α、β、γ、δ、ε和ζ:PLDα是磷脂酰肌醇4,5二磷酸(phosphatidylinositol4,5-bisphosphate;PIP2)-非依賴(lài)性的Z常見(jiàn)的植物PLD酶,而PLDβ和PLDγ是磷酸磷脂酰肌醇輔因子(polyphosphoinositidecofactor)依賴(lài)性;PLDγ在氨基酸序列同源性和生化特性上類(lèi)似于PLDβ;PLDδ為膜相關(guān)性,游離油酸(oleicacid)可以激活。PLD通過(guò)催化磷脂酰膽堿(phosphatidylcholine;PC)、磷脂酰甘油(phosphatidylglycerol;PG)、磷脂酰絲氨酸(phosphatidylserine;PS)和磷脂酰乙醇胺(phosphatidylethanolamine;PE)上磷酸二酯鍵(phosphodiester)的水解,產(chǎn)生磷脂酸和游離頭端基團(tuán)(freeheadgroup);磷脂酸作為信號(hào)分子,進(jìn)一步水解為二?;视?,與跨膜信號(hào)功能、亞細(xì)胞結(jié)構(gòu)質(zhì)膜的降解和重構(gòu)等有關(guān)。同時(shí)通過(guò)轉(zhuǎn)移磷脂基團(tuán)到原生醇類(lèi)的羥基上的磷脂轉(zhuǎn)移活性(transphosphatidylation)。PLD及其催化產(chǎn)物參與了植物組織對(duì)于環(huán)境應(yīng)急反應(yīng),例如創(chuàng)傷、寒冷、干旱缺水、鹽化高滲透壓等?;诘孜锪字D憠A和磷脂酰乙醇胺,在磷脂酰肌醇4,5二磷酸和油酸的輔助下,通過(guò)磷脂酶D的作用,水解產(chǎn)生為磷脂酸和各種基團(tuán)后,通過(guò)脂酶(lipase)、甘油-3-磷酸氧化酶(glycerol-3-phosphateoxidase)和過(guò)氧化酶(peroxidase)系統(tǒng),測(cè)定紅色產(chǎn)物醌亞胺染料(quinoneiminedye)的峰值變化(500nm波長(zhǎng)),來(lái)定量分析磷脂酶D的總水解活性。磷脂酶D連續(xù)循環(huán)反應(yīng)系統(tǒng)為:[詳細(xì)]
